Sermorelin
is a synthetic peptide analog of Growth Hormone-Releasing Hormone (GHRH). Specifically, it is composed of the first 29 amino acids of the naturally occurring 44-amino acid human GHRH (GHRH(1-29)-NH2). This fragment retains the full biological activity of the endogenous hormone in stimulating growth hormone (GH) release.
It is classified as a growth hormone secretagogue and serves as a research tool to specifically stimulate the pituitary's somatotropic cells via the GHRH receptor (GHRH-R).
Pituitary Receptor Activation: Sermorelin binds with high affinity to the GHRH receptor on somatotroph cells in the anterior pituitary gland.
Intracellular Signaling Cascade: Receptor binding activates a G-protein coupled pathway, leading to increased intracellular cAMP and calcium influx.
Pulsatile GH Secretion: This cascade triggers the synthesis and release of stored growth hormone into the systemic circulation. Crucially, Sermorelin stimulates a physiological, pulsatile pattern of GH release, mimicking the body's natural secretory bursts.
Feedback Loops: The subsequent rise in GH stimulates insulin-like growth factor 1 (IGF-1) production primarily in the liver, which then exerts negative feedback on further GH secretion at both the pituitary and hypothalamic levels.
